Recombinant Human Pegylated Interferon-alpha 2a - rHuPEG-IFN-a 2a

Description: Recombinant Human IFN-α 2a produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 165 amino acids and having a molecular mass of 19241 Dalton. The Interferon-a 2a gene was obtained from human leukocytes. rHuPEG- IFN-α 2a is manufactured by attaching a 40,000 dalton methoxypolyethylene glycol propionaldehyde (mPEG-ALD) to the N-terminal amino acid of IFN-α 2a having a total molecular mass of 59,241 dlaton.. The rHuIFN-α 2a is purified by proprietary chromatographic techniques.

Physical Appearance: Sterile Filtered clear liquid formulation.
Formulation: The protein 0.18mg/ml solution contains 8mg sodium chloride, 0.05mg tween 80, 0.05mg acetic acid and 2.62mg sodium acetate pH=6.
Stability: rHuPEG-IFN-α 2a should be stored between 4°C-8°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Purity: Greater than 98.0% as determined by both: (a) Analysis by RP-HPLC. (b) Anion-exchange FPLC. (c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Amino acid sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Cys-Asp-Leu-Pro-Gln, conforming to the sequence of native human IFN-α. N-terminal methionine has been completely removed enzymatically.
Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Biological Activity: ProSpec’s rHuPEG-IFN-α 2a is fully biologically active when compared to standard. The specific activity as determined in a viral resistance assay using bovine kidney MDBK cells was found to be 0.7 x 108.
Endotoxin: Less than 0.1 ng/µg (IEU/µg) of rHuPEG-IFN-α 2a.
Protein content: Protein quantitation was carried out by two independent methods: UV spectroscopy at 280 nm using the absorbency value of 0.924 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). Analysis by RP-HPLC, using a calibrated solution of IFN-α 2a as a Reference Standard.
